The immunoassay is intended for the qualitative detection of specific human IgG class antibodies to Toxoplasma gondii, Rubella virus, Cytomegalovirus and Herpes Simplex Virus (HSV-1 and HSV-2) in human serum.

Multiplex assay protocol

1. ADDITION OF SUSPENSION

A pre-diluted blood serum is mixed with a suspension of polystyrene beads. The beads are encoded with different amount of fluorescent dye. Antigens (Ag) of pathogens are chemically coupled to the bead’s surface.
 

2. BINDING REACTION

In the case of presence of IgG specific to pathogens Ag in serum, formation of complexes Ag – specific antibody occurs in the reaction mixture. For example:

Ag (CMV) – IgG;

Ag (Toxo) – IgG.

3. ADDITION OF CONJUGATE

The beads suspension is washed. Conjugate (Conj) is added to the washed microspheres. The conjugate consists of antibodies specific to human IgG and labeled with a fluorescent label (phycoerythrin).

4. COMPLEX FORMATION

In the reaction mixture Ag – specific antibody – Conj complexes are formed on the surface of the beads:

Ag (CMV) – IgG – Conj

Ag (Toxo) – IgG – Conj

Control bead  – Conj

5. ANALYSIS

The suspension is analyzed using any flow cytometer.

Features and benefits

The method of flow cytometry is used for analysis. The analysis can be performed on flow cytometers of any manufacturer.
